Abstract: 目的探讨茶多酚（TP）对草甘膦致小鼠睾丸支持细胞（SC）损伤的拮抗作用。方法原代培养小鼠睾丸支持细胞，采用FasL免疫细胞化学法鉴定支持细胞。建立草甘膦诱导支持细胞损伤模型，将细胞随机分为正常对照组、草甘膦损伤组（90btg／m1）、TP拮抗组（经10、20、40、80、160μg／mlTP预处理12h后，再用草甘膦作用细胞24h）。四噻唑蓝（MTT）法检测细胞存活率，倒置相差显微镜观察细胞形态学改变，原位末端标i5（TUNEL）法原位检测细胞凋亡，Hoechst33342荧光染色法观察细胞核形态改变，并检测细胞氧化损伤指标超氧化物歧化酶（SOD）、谷胱甘肽过氧化物酶（GSH—Px）、丙二醛（MDA）的变化。结果10～80μg／mlTP可拮抗草甘膦诱导的支持细胞损伤，提高支持细胞活力及SOD、GSH—Px活性，降低细胞凋亡指数及MDA含量，改善细胞萎缩变形、染色质浓集现象；160μg／ml时不能减轻草甘膦造成的细胞损伤。结论TP在一定浓度范围内对草甘膦诱导的支持细胞损伤具有拮抗作用。 Keywords: tea polyphenols, mouse, testicular support cell, glyphosate, Sertoli cells, mice, testis, rodent Citation: Hui, Y., Wen-hong, Z., Lia-hua, Z., Chun-fang, L. and Cheng-mei, J., 2013. The antagonistic effects of tea polyphenols on damage of mouse sertoli cells induced by glyphosate. Journal of Nutrition, 3.
