Abstract: 目的探讨茶多酚(TP)对草甘膦致小鼠睾丸支持细胞(SC)损伤的拮抗作用。方法原代培养小鼠睾丸支持细胞,采用FasL免疫细胞化学法鉴定支持细胞。建立草甘膦诱导支持细胞损伤模型,将细胞随机分为正常对照组、草甘膦损伤组(90btg/m1)、TP拮抗组(经10、20、40、80、160μg/mlTP预处理12h后,再用草甘膦作用细胞24h)。四噻唑蓝(MTT)法检测细胞存活率,倒置相差显微镜观察细胞形态学改变,原位末端标i5(TUNEL)法原位检测细胞凋亡,Hoechst33342荧光染色法观察细胞核形态改变,并检测细胞氧化损伤指标超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH—Px)、丙二醛(MDA)的变化。结果10~80μg/mlTP可拮抗草甘膦诱导的支持细胞损伤,提高支持细胞活力及SOD、GSH—Px活性,降低细胞凋亡指数及MDA含量,改善细胞萎缩变形、染色质浓集现象;160μg/ml时不能减轻草甘膦造成的细胞损伤。结论TP在一定浓度范围内对草甘膦诱导的支持细胞损伤具有拮抗作用。 Keywords: tea polyphenols, mouse, testicular support cell, glyphosate, Sertoli cells, mice, testis, rodent Citation: Hui, Y., Wen-hong, Z., Lia-hua, Z., Chun-fang, L. and Cheng-mei, J., 2013.
